To do so, I created an aggregate audio device on my Mac, named it Rocksmith USB Guitar Adapter, and simply used the Focusrite 2i2 instead of the Rocksmith Real Tone cable. Plug in any real guitar or bass and join over 1 million people who have learned with the award-winning Rocksmith method. Rocksmith 2014 Edition is the fastest way to learn guitar. With MME, I'm able to record the guitar while monitoring the drum track but a) the 30ms latency makes it pretty much impossible, and b) in any case it only records/plays on the left channel, regardless of how I adjust the input/output settings on the track itself as well as in Channel Mapping. I have successfully used Rocksmith with a Focusrite 2i2 interface from my MacBook. CAUTION: THIS GAME CANNOT BE PLAYED WITHOUT THE ROCKSMITH® REAL TONE CABLE. This cable is compatible with all versions of Rocksmith on all available platforms.
I've tried disabling 3.0 support in the bios, and no change. My problem is that my realtone cable will not be recognized, whether I put it in a USB 2.0 or 3.0 socket.

Rocksmith usually includes the Real Tone Cable in the box, except downloaded versions. The SSD holds windows and programs, while the HDD holds documents, music, and games, including Rocksmith. Oh, and not sure if it's supposed to be this way, but I notice that MME gives me two options under Audio Hardware: one for input and one for output, while WASAPI only gives me one. The Rocksmith Real Tone Cable is a unique 1/4'-to-USB cable that enables Rocksmith to detect and respond to your electric guitar or bass. Obviously I have a lot to learn about the basics of the process, but in all my searching through Adobe's how-to pages and YouTube tutorials, I haven't been able to get the setup to work properly. You can also use a dedicated inch male to ¼ inch male cable. Assuming you already own a guitar cable you will just need a 1/8 inch male to 1/4 female adapter which costs around 5 so you will be able to connect your guitar to the mic jack.

I've tried a variety of input/output settings on the two tracks, but so far have had no luck. This is the cheapest way to connect your electric guitar to a PC or laptop. Interestingly, even though it says "Not working," I was able to record a sample, but thus far have been unable to do so while monitoring the drum track.
To answer your question, yes, it does work in MME (I have no ASIO option), but the lowest latency is 30ms, inadequate for recording against the drum track that's already on there.
